Key points :
- EVT801 is a selective inhibitor of vascular endothelial growth factor 3 (VEGFR3). VEGFs and VEGFRs are well-validated cancer drug targets with multiple FDA-approved products, but the greater selectivity of EVT801 may result in better tolerability and less development of resistance to therapy.
- EVT801 was confirmed to be a potent and selective inhibitor of VEGFR3, with activity in the low nanomolar range. The drug was shown to inhibit the formation of lymphatic vessels in vitro, which is its intended primary mode of action.
- In vivo (animal) experiments showed EVT801 to be more effective than both pazopanib (Votrient®, Novartis) and sorafenib (Nexavar®, Bayer) in the tumour models under investigation.
- EVT801 combined with immune checkpoint inhibitors in mouse models of several tumour types showed strongly synergistic activity, with the combination performing better than either drug alone, and with some non-responders to immunotherapy becoming responsive after the addition of EVT801. Immune checkpoint inhibitors are widely used in many cancers, and the class includes drugs such as Keytruda® (pembrolizumab), Opdivo® (nivolumab), and Yervoy® (ipilimumab).

L'IUCT-Oncopole, centre régional de soin, de recherche et de formation en cancérologie regroupe à Toulouse
l’expertise de 1 800 professionnels sur un même site labellisé « Comprehensive Cancer Center ». Il combine
plusieurs installations cliniques de pointe pour le traitement du cancer avec une infrastructure de recherche de
classe mondiale, sur un campus intégré qui rassemble des parties prenantes publiques et privées, y compris des
partenaires industriels. L’IUCT-Oncopole, qui réunit l’Oncopole Claudius Regaud (ICR) et plusieurs équipes du CHU
de Toulouse, traite plus de 10 000 nouveaux patients chaque année, et plus d'un patient sur huit est inscrit dans
des études cliniques.
Grâce à l’équipe d’hématologie du CHU, l’IUCT-Oncopole est le centre de référence régional pour
l’oncohématologie et a inauguré l’immunothérapie personnalisée à Toulouse, en traitant ses premiers patients
par CAR-T cells dès 2019, en collaboration avec le réseau Onco-Occitanie.
